Abstract
Betulinol (Bet) is a bioactive pentacyclic triterpenoid with low water solubility. In this study, caffeic acid phenethyl ester (CAPE) and glycyrrhetinic acid (GA) were chosen as guest molecules to coassemble with Bet, and the characteristics of Bet-GA/CAPE coassemblies were investigated. The results show that the Bet-CAPE coassembly with a nanolamellar structure forms at a mass ratio of 7:3, while nanosphere Bet-GA forms at 8:2, with the latter having the best storage stability, thermal stability, pH stability, salt stability, and photostability. After coassembly, the solubility of Bet was greatly enhanced, 78.67 ± 4.16% for Bet-CAPE at 7:3 and 59.33 ± 1.15% for Bet-GA at 8:2. In addition, a synergistic antioxidant effect from Bet-CAPE and Bet-GA coassemblies was found. However, the release of Bet from the Bet-CAPE coassembly was different from that of the Bet-GA coassembly. The coassembly technique provides a new idea for expanding the application of Bet, CAPE, and GA.
